pBAD18 Plasmid
pBAD18 is a low-copy, arabinose-inducible expression vector designed for tightly regulated gene expression in Escherichia coli. It features the PBAD promoter, which is activated by L-arabinose in the presence of the AraC regulatory protein, enabling precise control over expression levels with minimal basal activity. This makes pBAD18 particularly well-suited for expressing genes that are toxic, unstable, or otherwise challenging to express constitutively.
The plasmid carries an Ampicillin resistance marker (AmpR) for antibiotic selection and replicates using the p15A origin of replication, which supports a low-copy number (~10–12 copies per cell). This origin is compatible with ColE1-based plasmids, allowing for co-transformation strategies in dual-plasmid systems.
pBAD18 is part of the classic arabinose-inducible expression system family, offering tighter expression control with reduced plasmid burden. For higher yield expression under similar regulation, pBAD24 is a commonly used alternative based on a ColE1 origin.
