CV-COLTRC (Eq. pTrc99a)
CV-COLTRC (Eq. pTrc99a)
CV-COLTRC has a sequence identical to the pTrc99A expression vector. This medium-copy plasmid carries an ampicillin resistance marker and a ColE1-derived origin, and features the hybrid trc promoter for IPTG-inducible expression in E. coli. It is a versatile backbone for regulated protein expression and general cloning applications.

Background
pTrc99A was introduced as part of the trc promoter system, a hybrid promoter combining elements of the trp and lac promoters to enable strong, IPTG-inducible expression in E. coli. This design provided tighter regulation and higher expression than earlier lac-based systems. CV-COLTRC preserves the exact sequence of pTrc99A, ensuring compatibility with the established literature and experimental protocols. The original description can be found in: Amann, E., Brosius, J., & Ptashne, M. (1988). Vectors bearing a hybrid trp-lac promoter useful for regulated expression of cloned genes in E. coli. Gene, 69(2), 301–315.
Features
- Antibiotic selection: Ampicillin (bla)
- Origin of replication: ColE1-derived, medium copy (~15–20/cell)
- Size: ~4.6 kb
- Promoter system: Hybrid trc promoter (trp–lac), IPTG-inducible
- MCS: Multiple cloning site downstream of trc promoter
- Regulatory elements: lacIᵠ for tighter repression of basal expression
Applications
- Regulated protein expression in E. coli
- Cloning and expression of recombinant proteins
- Functional studies requiring adjustable induction strength
- Educational use for teaching inducible expression systems
Limitations
- Requires IPTG induction; basal expression may still occur in some strains
- Medium copy number yields less DNA than pUC-derived vectors
- Limited to E. coli expression; not suitable for other hosts
